Thermo Fisher Scientific is seeking partners and opportunities to develop and commercialize novel technologies to detect protein:protein, protein:nucleic acid, and/or

protein:small molecule interactions. 



Protein interactions are important in drug discovery, studying cell signaling mechanisms, and understanding protein function and structure. Often more than one approach or methodology is necessary to validate an interaction.


Key Success Criteria

The successful technology should meet some or all of the following criteria:

  • Detect transient interactions
  • Capable of detecting multiprotein complexes
  • Amenable to use in vitro and in cultured cells
  • Able to quantify the interaction (frequency and/or affinity)
  • Identify subcellular location of interaction
  • Detect posttranslational modifications
  • Reproducible
  • Minimizes operator hands-on time


Possible Approaches
  • Stabilization of protein complexes prior to isolation
  • Detection of endogenous protein complexes
  • Over-expression systems employing detectors, unique tags, or affinity handles
  • Label-free detection


Approaches not of interest
  • Yeast-2-hybrid and yeast-1-hybrid
  • Immunoprecipitation and co-immunoprecipitation
  • Phage display
  • Gel shift assay
